国家重点保护植物赤水蕈树的遗传多样性
Genetic diversity of national key protected plant Altingia multinervis
摘要
Abstract
[Objective]The genetic diversity of 10 wild populations of Altingia multinervis W.C.Cheng,a national secondary key pro-tected plant in Guizhou Province,was analyzed to provide reference for the protection and research of rare and endangered plants.[Method]Inter-simple sequence repeat(ISSR)molecular markers were used to analyze the genetic diversity of 38 samples from 10 wild A.multinervis populations.[Result]A total of 188 clear bands were obtained via PCR amplification using 10 polymorphic primers,including 159 polymorphic bands,which accounted for 84.57%of the total.The A.multinervis populations had a Nei's diversity index of 0.267 4 and Shannon information index of 0.407 2.Considering a genetic differentiation coefficient of 0.4 and mean gene flow of 0.654,significant genetic differentiation was predicted to have occured among the A.multinervis populations.The A.multinervis populations had genetic consistency of 0.790 2~0.932 6 and genetic distance of 0.069 8~0.235 4,and Mantel test further verified that there was no obvious geographical trend in genetic diversity among the populations.Lastly,the 38 samples from 10 wild A.multinervis populations were divided into 2 categories by UPGMA cluster analysis of the genetic relationship.[Conclusion]The genetic variation of wild A.multinervis was at a medium level,and the genetic diversity of different A.multinervis populations was quite different.关键词
