毛竹miR156靶向SPL19A调节植物年龄发育
Moso bamboo miR156 targeting SPL19A to regulate plant age development
摘要
Abstract
[Objective]This study focused on the role of miR156(PhemiR156)and its target gene SPL(PheSPL)in regulating age development of moso bamboo(Phyllostachys edulis),to provide a theoretical basis for research on its flowering mechanism and ge-netic improvement.[Method]PhemiR156 and PheSPL genes were screened by homologous sequence alignment,and then analyzed for their sequences,structures,physicochemical properties and expression patterns with bamboo transcriptome data.Arabidopsis transgenic lines overexpressing PheMIR156 and PheSPL were constructed,and were analyzed for age development phenotypes.[Re-sult]Fourteen PhemiR156 precursor genes and 36 PheSPL homologous genes were identified in bamboo.Among these PheSPL genes,17 genes have miR156-binding sites,and 32 genes encoded proteins containing SBP domain.The expression level of Phe-miR156 were decreased with bamboo age,while that of PheSPL was higher during the reproductive development stage.Overexpres-sion of PheMIR156H significantly inhibited the age development of Arabidopsis,whereas the overexpression of PheSPL19A led to an earlier transition to adulthood.[Conclusion]PheMIR156H and its target gene PheSPL19A inhibited and promoted the age develop-ment of bamboo,respectively.关键词
