3株FAdV-Ⅰ在LMH细胞上的培养特性及不同病毒传代方法的比较
Cultivation characteristics of 3 FAdV-Ⅰ strains in LMH cells and comparison of different methods for virus passage

Abstract
[Objective]To provide a better passage method for fowl adenovirus group I(FAdV-Ⅰ),the culture characteristics of different FAdV-Ⅰ strains in leghorn male hepatoma(LMH)cells as well as merits and demerits of different virus passage methods were investigated.[Method]Three FAdV-Ⅰstrains(CELO,GY and TR22-CK8)were inoculated into LMH cells for virus passage and determination of passage number by observing its cytopathic effect.Then the virus was harvested by conventional method and freeze-thaw subculture,and the 50%tissue culture infective dose(TCID50)was determined.Different loads of FAdV-Ⅰwere added to chicken embryo allantoic fluids infected with infectious bronchitis virus(IBV),and the FAdV-Ⅰ load in the IBV live vaccine was detected by indirect immunofluorescence assay(IFA)after propagated by serial passage or freeze-thaw subculture.[Result]All the 3 FAdV-Ⅰ strains can induce the characteristic cytopathic effects of avian adenovirus on LMH cells,but with different timings and degrees.Passage with virus would worsen the cytopathic effects,but no more than 3 serial passages.The virus loads propagated by freeze-thaw subculture were higher,and the cells were less likely to detach.[Conclusion]Different FAdV-Ⅰstrains had various adaptability and culture characteristics on LMH cells,and freeze-thaw subculture was more suitable for the propagation of FAdV-Ⅰ.关键词
