A new colorimetric detection method for beer spoilage lactic acid bacteria based on non-woven fabric composite and improved MRS liquid culture medium

Abstract
As beer spoilage lactic acid bacteria(LAB)can lead to beer spoilage,the detection of LAB is an inev-itable requirement for beer industry.This paper reported a new colorimetric detection method for beer spoilage LAB based on non-woven fabric composite and improved De Man,Rogosa and Sharp medium(MRS)liquid medium.The method can detect beer spoilage LAB(1 CFU/15 mL)within 48 h methyl red and bromocresol green was used as the indicator of beer spoilage LAB.If the filter membrane changes from green to yellow,it is positive for spoilage bacteria;if not,it is negative.Deionized water was replaced with 75%beer mixture in the improved MRS medium for simulating the native environment of spoilage bacteria LAB.And 50 U/mL catalase was used to activate beer spoilage bacteria in low metabolism state.Membrane filtration method was also used to enrich beer spoilage LAB from low-concentration beer.Hydrophilic non-woven fabric can adsorb liquid medi-um to provide efficient nutrition supply.The method is low in cost,high in efficiency and easy to operate to use in the factory.关键词
