黄芩素通过抑制NLRP3炎性通路改善卵巢刺激后的子宫容受性
Baicalein improves uterine receptivity after ovarian stimulation by inhibiting the NLRP3 inflammasome signaling pathway

Abstract
This study aimed to investigate the effects and potential mechanisms of baicalein in alleviating embryo loss induced by ovarian stimulation.Female ICR mice aged 6-8 weeks were randomly divided into 5 groups.Group 1,the natural cycles(NC)group;group 2,the ovarian stimulated(OS)group;group 3,the baicalein treatment(OS+Bai)(10 mg/kg)group;group 4,the baicalein treatment(OS+Bai)(25 mg/kg)group;group 5,the baicalein treatment(OS+Bai,100 mg/kg)group.First,the optimal dose of baicalein for ovarian stimulated mice was screened.Subsequently,Western blot,immunofluorescence and enzyme-linked immunosorbent assay were used to determine the expression levels of NOD-like receptor protein 3(NLRP3),apoptosis-associated speck-like protein containing a CARD(ASC),cysteine-aspartic acid protease-1(Caspase-1),gasdermin-D(GSDMD),IL-18,and IL-1β in the uterus.RT-qPCR was employed to detect the mRNA expression levels of uterine receptivity markers leukemia inhibitory factor(Lif)and mucin 1(Muc1).Furthermore,the birth weight and the average number of pups per litter produced were measured.The results showed that,compared with group 1,the embryo implantation rate in group 2 was significantly reduced(P<0.05).Among the three baicalin-treated groups,group 3 exhibited the highest embryo implantation rate.The levels of NLRP3,ASC,Cleaved-CASP-1,GSDMD,IL-18 and IL-1β in group 3 were significantly lower than those in group 2(P<0.05).Moreover,the average birth weight and the average number of pups per litter in group 3 were significantly higher than those in group 2(P<0.05).In conclusion,baicalein improves the uterine receptivity decline induced by ovarian stimulation and thus ameliorates the pregnancy outcome by inhibiting the NLRP3 inflammasome activation.关键词
