干旱环境胁迫后小鼠肺免疫稳态的变化及生脉散作用特点研究
Changes of Pulmonary Immune Homeostasis Imbalance in Mice after Drought Environmental Stress and Action Characteristics of Shengmai-san

Abstract
Objective:To observe the changes of pulmonary immune homeostasis imbalance in mice under drought stress and the action characteristics of Shengmai-san.Methods:Thirty-two healthy C57BL/6 mice were randomly divided into normal environment control group(NE group),NE plus Shengmai-san group(NEt group),arid environment control group(AE group),and AE plus Shengmai-san group(AEt group),with 8 mice in each group.The NE and NEt groups were placed in a normal temperature and hu-midity environment,while the AE and AEt groups were exposed to an artificial arid environment(with dryness pathogen intervention)in a climate chamber.The NEt and AEt groups were intragastrically administered with Shengmai-san[12.48 g/(kg·d)],and the NE and AE groups were intragastrically administered with an equal volume of normal saline for 48 consecutive days.After intervention,open-field test and fatigue swimming test were used to analyze the exploratory behavior and physical fitness,HE staining to observe pathological changes of lung tissues,Western blot to detect the relative expressions of Toll-like receptor 2(TLR2),myeloid differentia-tion primary response protein 88(MyD88),phosphorylated MyD88(p-MyD88),nuclear factor κB(NF-κB),and lung surfactant proteins A and D(SP-A,SP-D),and a biochemical analyzer to measure alanine aminotransferase(ALT),aspartate aminotransferase(AST),blood urea nitrogen(BUN),and creatinine(Cr).Results:Compared with the NE group,the AE group showed decreased daily food intake and water intake(P<0.05),decrease body weigh gain(P<0.05),elevated AST level(P<0.05),mild thickening of alveolar septa with alveolar collapse under light microscopy,up-regulated lung TLR2 expression(P<0.05),and down-regulated lung SP-D expression(P<0.05),and the AEt group showed decreased serum BUN level(P<0.05);Compared with the AE group,the AEt group showed decreased daily food intake and water intake(P<0.05),decreased exploratory ability(P<0.05),relatively complete alveolar structure under light microscopy,down-regulated lung TLR2 and p-MyD88 expressions(P<0.05),and up-regulated lung SP-D expression(P<0.05).Conclusion:Long-term drought stress did not disrupt the balance between pro-inflammatory and anti-inflammatory responses in murine pulmonary tissues,but impaired local innate immune barrier function by down-regulating pulmonary SP-D expression and compromising alveolar structural integrity.Shengmai-san can preserved local pulmonary immune homeostasis by modulating the TLR2/MyD88/NF-κB pathway and maintaining pulmonary SP-D expression.关键词
