电针足三里穴抑制自噬减轻APAP诱导药物性肝纤维化的研究
Electroacupuncture at Zusanli(ST36)Inhibiting Autophagy to Alleviate APAP-induced Hepatic Fibrosis

Abstract
Objective:To observe the effect of electroacupuncture at Zusanli(ST36)on APAP-induced hepatic fibrosis,and to explore its possible mechanism from the perspective of inhibiting autophagy via the phosphatidylinositol 3-kinase(PI3K)/mammalian target of rapamycin(mTOR)/transcription factor EB(TFEB)pathway.Methods:Thirty male SPF-grade SD rats were selected and randomly divided into three groups:control group(intragastric administration of normal saline),APAP group(intragastric administra-tion of APAP solution),and APAP-electroacupuncture group(immediate electroacupuncture intervention after APAP intragastric ad-ministration,bilateral Zusanli electroacupuncture stimulation for 30 minutes daily,dense-sparse wave frequency 20 Hz/100 Hz,inten-sity 1-2 mA,continuous intervention for 6 weeks).Masson staining was used to evaluate the fibrotic status of liver tissues;Western blot to detect the expression levels of p-PI3K,p-mTOR,p-TFEB,the spliced form of X-box binding protein 1(XBP1s),TFEB,autoph-agy-related protein 1(Beclin-1),and autophagy-related gene 5(ATG5)in liver tissues;and immunofluorescence to detect the fluo-rescence intensity of p-PI3K and TFEB.Results:Compared with the control group,the APAP group showed hepatic fibrosis,and the expressions of p-PI3K,p-mTOR,p-TFEB,XBP1s,TFEB,Beclin-1,and ATG5 were all increased(P<0.05);Compared with the APAP group,the APAP-electroacupuncture group showed a reduced degree of hepatic fibrosis(P<0.05),increased expressions of p-PI3K,p-mTOR,and p-TFEB(P<0.05),and decreased expressions of XBP1s,TFEB,Beclin-1,and ATG5(P<0.05).Conclu-sion:Electroacupuncture at Zusanli may effectively reduce autophagy caused by APAP-induced liver injury by activating the p-PI3K/mTOR/TFEB signaling pathway,thereby alleviating the fibrotic status of hepatocytes.关键词
