络通纤溶饮通过TGF-β1/Smads途径减轻特发性肺纤维化大鼠肺组织损伤的研究
Luotong Xianrong Decoction Alleviating Pulmonary Tissue Injury in Rats with Idiopathic Pulmonary Fibrosis via TGF-β1/Smads Pathway

Abstract
Objective:To observe the effect of Luotong Xianrong Decoction on the transforming growth factor-β1(TGF-β1)/Smads signaling pathway in lung tissues of rats with idiopathic pulmonary fibrosis(IPF),and to explore the possible mechanism of this formula in alleviating lung tissue injury in IPF rats.Methods:Thirty-two rats were randomly divided into sham operation group,model group,Luotong Xianrong Decoction group,and nintedanib group.Except sham operation guoup the IPF animal models of other groups were induced by tracheal instillation of bleomycin.On the second day after modeling,the Luotong Xianrong Decoction group was intra-gastrically administered with Luotong Xianrong Decoction(15.18 g/kg),the nintedanib group was intragastrically administered with nintedanib aqueous solution(0.1 g/kg),and the sham operation group and model group were given normal saline(10 mL/kg),once a day for 28 consecutive days.After lung function testing,lung tissues were harvested.HE and Masson staining were used to observe pathological morphological changes of the lung.Hydroxyproline(HYP)content was determined by alkali hydrolysis method.The ex-pression levels of TGF-β1,its type I receptor TGF-βRⅠ,p-Smad3,and p-Smad7 were detected by Western blot.Results:Compared with the sham operation group,the model group showed significant impairment of lung function and lung tissue injury,increased HYP content,and up-regulated protein expressions of TGF-β1,TGF-βRⅠ,and p-Smad3,as well as down-regulated p-Smad7 protein ex-pression(P<0.05);Compared with the model group,the Luotong Xianrong Decoction group exhibited alleviated lung function impair-ment and lung tissue injury,decreased HYP content,and down-regulated expressions of TGF-β1,TGF-βRⅠ,and p-Smad3,along with up-regulated p-Smad7 expression(P<0.05).Conclusion:Luotong Xianrong Decoction can significantly alleviate lung tissue inju-ry and improve lung function in IPF rats,exerting a protective effect against IPF.Its mechanism may be realized by inhibiting the TGF-β1/Smads signaling pathway.关键词
