基于肠道菌群探讨宣痹汤对高尿酸血症大鼠的作用研究
Mechanism of Xuanbi Decoction on rats with hyperuricemia based on intestinal flora

Abstract
Objective To explore the effect of Xuanbi Decoction on rats with hyperuricemia(HUA)based on intestinal flora.Methods Sixty SPF Wistar rats were randomly divided into a control group,a model group,an allopurinol group,and low-,medium-,and high-dose groups of Xuanbi Decoction using a random number table,with 10 rats in each group.On the 21st day,rat feces were collected and analyzed for differences in gut microbiota composition and metabolic pathways using the metagenomic sequencing technology,and the level of serum uric acid and the activities of xanthine oxidase and adenosine deaminase in them were measured using a reagent kit,while the pathological changes of rat small intestine tissue were observed by HE staining.Results Compared with the control group,the model group showed a significant increase in the level of blood uric acid,indicating a successful modeling,while the results of HE staining showed that the small intestinal villi became shorter.Compared with the model group,the levels of serum uric acid,xanthine oxidase,and the adenosine deaminase activities of the three groups of Xuanbi Decoction were all decreased(all P<0.05).The results of metagenomic sequencing showed that the Chao1 indexes of model rats were significantly increased after the intervention with Xuanbi Decoction(P<0.05).Beta diversity showed significant changes in the microbial composition between the model group and the high-dose Xuanbi Decoction group,with the latter being able to reverse the microbial composition of the model rats to some extent.KEGG enrichment functional analysis showed that the high-dose Xuanbi Decoction group up-regulated lipid metabolism and folded classification degradation(P<0.05),and down-regulated the biodegradation metabolism of xenobiotics,terpenoids,and polyketides(P<0.05).Conclusion Xuanbi Decoction can reduce the level of uric acid,inhibit the activities of ADA and XOD,and improve the pathological changes in the small intestines of HUA rats.Its mechanism of action may be related to its increasing the abundance of beneficial intestinal flora composition.关键词
