茵芍二黄汤改善小鼠免疫性肝炎的机制研究
Study on the effect of Yinshao Erhuang Decoction on immune hepatitis in mice based on regulating the Treg/Th17 imbalance

Abstract
Objective To observe the effect of Yinshao Erhuang Decoction on immune hepatitis in mice and explore its potential mechanism.Methods A total of 48 mice were randomly divided into a model group,Chinese medicine high-,medium-,and low-dose(53,26.5,13.25 g/kg)groups,a blank group,and a western medicine control group,and they were intervened by gavage continuously for 4 weeks.Concanavalin A(ConA)was injected into the tail vein once a week to induce an autoimmune hepatitis model,with the model constructed after 4 weeks.The liver and spleen indexes,serum ALT and AST levels,and IL-10 and IL-17A levels were measured.HE staining was used to observe the pathological conditions of liver tissue,and Western blot was used to detect the protein expression of Foxp3 and nuclear orphan receptor(RORγt)in liver tissue.Results Compared with the blank group,the liver and spleen indexes,ALT,AST and IL-17A levels were generally increased in the model group,while the IL-10 level was significantly decreased,the liver injury was obvious,and the protein expression of Foxp3 was decreased,while the RORγt protein expression was increased.Compared with the model group,the liver and spleen indexes,AST,ALT and IL-17A levels were decreased in the high-and medium-dose groups and the western control group,while the IL-10 levels were increased,and the liver injury was alleviated,and the Foxp3 protein expression was increased,while the RORγt protein expression was decreased.Conclusion Yinshao Erhuang Decoction has a certain protective effect on immune hepatitis induced by ConA in mice,and its mechanism may be related to regulating the Treg/Th17 imbalance.关键词
