摘要

Abstract

Cellulase is widely used in many industries,such as food,paper making,textiles and animal feed.However,the activity of the enzyme is easily decreased and difficult to be recycled,resulting in the increase in the cost and inability to large-scale application.Taking the enzyme activity fixation ratio as the objective,cellu-lase was immobilized by embedding-crosslinking,using sodium alginate-arabic gum as a composite carrier,glut-araldehyde-ethylene glycol glycidyl ether as a composite crosslinking agent.The effects of single factors,consis-ting of the amount of enzyme,the ratio of crosslinking agents,the ratio of carriers,immobilization time,and the concentration of CaCl2,on the immobilization results were investigated,and an orthogonal experiment was de-signed to optimize the immobilization technology.The results show that the enzyme activity fixation ratio of the immobilized cellulase is 99.60%under the conditions of the enzyme amount of 15 mL,a cross-linking agent ratio of 2∶3,the carrier ratio of 2.0∶2.5,immobilization time of 2.5 hours,accompanied by CaCl2 concentra-tion of 1.0%.The most suitable technological conditions lay the foundation for the large-scale application of cellulase.
