Abstract
The antioxidant and hypoglycemic activities of Dendrobium chrysanthum flowers with different polarity extracts were studied,and the types of inhibitory effects of the flower's extracts on α-amylase and α-glucosidase were explored to assess the potential of D.chrysanthum flowers for antioxidant and hypoglycemic food and drug applications.D.chrysanthum fresh flowers were extracted by 95%ethanol.The extract was successively extracted with organic solvents of different polarities to obtain petroleum ether extract(DC1),ethyl acetate extract(DC2),n-butanol extract(DC3),95%ethanol extract aqueous phase(DC4),and the residue was extracted by 65%ethanol to obtain DC5.Antioxidant activity was evaluated using DPPH·,·ABTS+and·OH radical scavenging experiments,and antihypoglycemic effect was determined using α-amylase andα-glucosidase.Further study of the type of enzyme inhibition was conducted through enzymatic kinetic methods and Line weaver Burk curves.The experimental results showed that the antioxidant activity of different extracts from D.chrysanthum flowers were weak,but they all had certain α-amylase and α-glucosidase inhibitory activities.The inhibitory activities of α-amylase was as follows:DC1>DC2>DC5>DC4>DC3.The half inhibitory mass concentration(IC50)of DC1 and DC5 on α-amylase inhibition activity were(116.87±6.27)and(107.15±4.15)mg/L,respectively,indicating mixed inhibition and anti-competitive inhibition types.The α-glucosidase inhibitory activity was as follows:DC2>DC4>DC1>DC5>DC3.Compared with the other four extracts,DC2 exhibited much greater inhibitory activity,with IC50 of(614.24±55.60)mg/L,indicating competitive inhibition.The research results indicated that the development and utilization of D.chrysanthum flowers had a great potential as a hypoglycemic food and drug.关键词
