白藜芦醇对LPS诱导的HGF细胞炎症反应、凋亡和细胞损伤的抑制作用及其机制研究
Study on the inhibitory effect of resveratrol on LPS-induced inflammation,apoptosis and cell damage in HGF cells and its mechanism

Abstract
Objective:To investigate whether resveratrol alleviates periodontitis through FGFR1/TLR4/NLRP3 in-flammatome pathway and its mechanism.Methods:Normal gingival fibroblasts(HGF)were cultured and randomly divided in-to Control group,LPS group,LPS+RES(12.5,25,50 μmol/L)group,LPS+PD166866 group and LPS+25 μmol/L RES+PD166866 group.In the LPS and resveratrol co-culture group,HGF cells were treated with different concentrations of RES for 24 h,and then stimulated with or without 5 μg/mL LPS for 48 h.HGF cells were treated with 10 μmol/L FGFR1 inhibitor PD166866 for 24 h or with 25 μmol/L RES,and then stimulated with 5 μg/niL LPS for 48 h.Cell viability was measured by CCK8 assay.LDH assay kit was used to evaluate LDH release,and Western blot was used to detect the protein expression lev-els of IL-6,IL-8,Bax,Bcl-2,FGFR1,TLR4 and NLRP3.Results:Compared with Control group,cell viability was decreased,LDH release was increased,protein expression levels of IL-6,IL-8,Bax,FGFR1,TLR4 and NLRP3 were significantly in-creased in LPS group,while the protein expression level of Bcl-2 was significantly decreased(P<0.01).Compared with the LPS group,the LPS+RES(12.5,25,and 50 μmol/L)group showed increased cell viability,decreased LDH release,and sig-nificantly decreased levels of IL-6,IL-8,Bax,FGFR1,TLR4,and NLRP3 protein expression,and significantly increased levels of Bcl-2 protein expression in a dose-dependent manner.Compared with LPS+25 μmol/L RES group,the cell viability of LPS+PD166866 group and LPS+25 μmol/L RES+PD166866 group was significantly decreased,and the LDH release was significantly increased.The protein expression levels of IL-6,IL-8,Bax were significantly increased.Conclusion:Resveratrol can inhibit LPS-induced inflammation,apoptosis and cell damage in HGF cells,and may regulate intracellular inflammation by targeting FGFR1 signaling pathway.关键词
