基于UV胶光固化制备馆藏陈列细菌平板标本的新方法
A Novel UV-Photocuring Adhesive Method for Long-Term Preservation of Bacterial Culture Plates for Exhibition

Abstract
Due to the limitations of the tfabrication methods by the raditional bacterial plate specimen in terms of light transmit-tance,whitening,and long-term room temperature preservation,these methods are unsuitable for museum exhibitions.In this study,a novel specimen preservation method of the bacterial plate was developed based on UV-curable adhesive,using Escherichia coli and Planomicrobium okeanokoites as representative bacterial models.By optimizing the culture medium composition and modifying the UV adhesive,the study achieved long-term room temperature conservation of the specimens.The results demonstrated that the optimized culture medium significantly enhanced specimen brightness(L*).In the E.coli and P.okeanokoites systems,the addition of low-salt or anti-crystallization agents increased L* from 39.36 to 40.75(E.coli)and from 38.15 to 40.35(P.okeanokoites),with total color differences(ΔE*)of 0.82 and 0.76,respectively,meeting the requirements for visual fidelity(ΔE*<3.0).Furthermore,benzotri-azole(BTA)-modified UV adhesive effectively reduced the yellow shift index(Δb*)by 79.3%,with a total color difference(ΔE*)of 1.50,thereby suppressing UV adhesive yellowing.This study employs a synergistic strategy combining low-salt culture medium opti-mization,anti-crystallization culture medium improvement,and BTA-modified UV adhesive,significantly enhancing specimen trans-parency and optical stability to meet the visual requirements of museum exhibits.关键词
