摘要

Abstract

Objective:To construct a radiation-induced positive feedback loop cArGs-LuxR/luxI and utilize the X-ray spatiotemporal effect to obtain targeting tumors with sustained high expression of p53,achieving radiotherapy sensitization.Methods:Using the retroviral vector Lvool as the backbone,reporter gene vectors(pE6-EGFP,pLuxI-luxR-EGFP,pE6-LuxI-luxR-EGFP)and p53 expression vector(pE6-p53,pLuxI-luxR-p53,pE6-LuxI-luxR-p53)were constructed.Transfect H1299 and A549 cell lines separately,administer different doses of X-ray irradiation,and observe the intensity of EGFP expression.Western blotting was used to detect p53 expres-sion.Flow cytometry was used to detect cell apoptosis,while cell cloning experiments,comet electrophoresis,and other ex-periments were used to detect the radiosensitizing effect of radiotherapy.Observe tumor inhibition effect in nude mouse tumor experiments,and immunohistochemical detection of p53 expression in tumor and adjacent tissues.Results:After sequencing verification,the vectors described above were successfully constructed.The pE6-LuxI-LuxR-p53 vector exhibits radiosensitivi-ty and maintains sustained high expression of p53,with the strongest expression observed at 4 Gy.The radiotherapy sensitiza-tion ratio is 1.38.In transplanted tumors,the pE6-LuxI-luxR-p53+IR treatment group showed the strongest anti-tumor effect(P<0.01).The expression level of p53 increased in irradiated wild tumor tissues(P<0.05),while there was no p53 expres-sion in the adjacent tissues of the wild tumor.Conclusion:Under irradiation induction,the pE6-LuxI-LuxR-p53 vector tar-gets the high expression of p53 and has significant radiation sensitization and tumor inhibition effects.
