Investigating the Effects of All-Trans Retinoic Acid on HBL-100 Cells Using RNA-Seq Technology

Abstract
Objective:This study aims to explore the impact of all-trans retinoic acid(ATRA)on gene expression in the breast epithelial cell line HBL-100,with the goal of elucidating the potential mechanisms of ATRA underlying breast physiol-ogy and pathology.Methods:Gene expression changes in HBL-100 cells before and after ATRA treatment were analyzed u-sing RNA-Seq technology.Differentially expressed genes(DEGs)were identified with the criteria of|log2FC|≥1 and P<0.05.The DAVID online tool was employed for Gene Ontology(GO)enrichment analysis and KEGG pathway analysis to de-termine the biological functions and signaling pathways of the DEGs.Additionally,the STRING database was used to construct a protein-protein interaction(PPI)network,and key hub genes were identified using the cytoHubba plugin.Results:A total of 580 DEGs were identified,with 303 up-regulated and 277 downregulated.Upregulated genes were mainly involved in processes such as viral defense,immune response,and apoptosis,while downregulated genes were associated with hormone signaling,cell adhesion,and metabolic pathways.KEGG analysis of upregulated genes showed enrichment in immune signaling pathways,including NOD,TOLL,and RIG-I signaling pathways,while downregulated genes were significantly enriched in pathways related to endoplasmic retic-ulum protein processing,PI3K-Akt signaling pathway,and metabolic processes.PPI network analysis identified 10 upregu-lated hub genes,including STAT1,CXCL10,and DDX58,as well as 10 downregulated hub genes,such as SPP1,APOE,and CANX,which may play crucial regulatory roles under ATRA treatment.Conclusion:This study demonstrates that ATRA significantly affects the gene expression profile of HBL-100 cells,particularly genes related to immune response and cell sig-naling.These findings provide new insights into the role of ATRA in breast epithelial cells and lay the groundwork for further exploration of ATRA's potential in breast cancer prevention and treatment.关键词
