Abstract

The experiment aimed to screen for lactic acid bacteria from the intestinal tract of broiler chickens that meet the criteria for probiotic characteristics and have antibacterial activity.Four strains of lactic acid bacteria(Lab17,Lab23,Lab33,Lab65)were isolated and purified from the intestinal contents of broiler chickens through selective cultivation,and identified by 16S RNA sequence alignment and phylogenetic tree analysis.Screening strains that meet the basic standards of probiotics through growth tests,acid resistance tests,and bile salt tolerance tests.Evaluate the in vitro probiotic characteristics of the strain through relevant indicators such as antibacterial tests,acid production performance tests,and drug sensitivity tests.The results showed that Lab23,Lab33,and Lab65 exhibited good growth ability in MRS medium,Lab17,Lab23,and Lab65 had strong acid resistance,Lab17,Lab33,and Lab65 had strong resistance to bile salts.The antibacterial test results showed that the fermentation supernatant of Lab65 had good inhibitory effects on enterotoxigenic Escherichia coli,Salmonella typhimurium,and Salmonella enterica.The acid production capacity test results showed that after 24 hours of cultivation,the pH value of Lab65 strain remained stable around 3.4.The drug sensitivity test results showed that the Lab65 strain was only resistant to ceftriaxone among the 16 antibiotics tested.The results show that the Lactobacillus rhamnosus Lab65 screened in this study meets the probiotic characteristic standards,has good gastrointestinal environmental tolerance,significant antibacterial activity,stable acid production ability,and antibiotic resistance spectrum meets the safety standards for feed probiotics.It has the potential to be developed as a probiotic for poultry feed.
