Sequence analysis of the rbcL barcode in Cibotii Rhizoma and its adulterants

Abstract
Objective Cibotii Rhizoma is a widely used Chinese medicinal.Due to its surging demand,its adulterants and counterfeits have proliferated in the market.Its commercial products are sun-dried rhizome slices,which are challenging to accurately identify using traditional methods.Therefore,there is an urgent need to establish a simple and rapid DNA barcoding system for its authentication.Methods The rbcL sequences of the genuine species Cibotium barometz and its adulterants(Adiantum raddianum,Woodwardia unigem-mata,Phlebodium aureum,and Pteris wallichiana)were amplified,sequenced,genetically analyzed and phylogenetic tree was construc-ted.Results Genetic distances was calculated based on the K2P model,which revealed that the intraspecific genetic distances of C.barometz were significantly smaller than those between C.barometz and its adulterants.Phylogenetic tree results demonstrated that the rbcL of C.barometz formed a distinct clade,demonstrating significant genetic divergence between species.Conclusion The rbcL se-quence can serve as an DNA barcode for successful identification of C.barometz,providing a theoretical and practical foundation for en-suring the quality of this medicinal material.关键词
