祛脂愈肝汤对非酒精性脂肪性肝炎细胞脂质代谢及IL-17表达的影响
Effect of Quzhi Yugan Decoction(祛脂愈肝汤)on lipid metabolism and IL-17 expression in non-alcoholic steatohepatitis cells

Abstract
Objective To observe the effect of QuzhiYugan Decoction(祛脂愈肝汤,QZYGD)on lipid metabolism and IL-17 expres-sion in a non-alcoholic steatohepatitis(NASH)cell model.Methods The NASH model was established by 1mmol/L free fatty acid(FFA)-induced mixture.Cells were divided into the following groups:the blank group,the model group,the silence group(100 μl blank rabbit serum),low/medium/high-dose QZYGD groups(100 μl of 5%,10%,and 15%medicated serum,respectively),and the polyene phosphatidylcholine group(100 μl of 10%medicated serum).Oil Red O staining was used to observe staining conditions of lipid droplets in NASH cells.RT-qPCR and Western blot(WB)were used to detect the expression of IL-17 mRNA and protein.Re-sults Cells in the blank control group exhibited clear structures,blue nuclei,and no significant orange-red lipid droplets.The model group showed numerous orange-red lipid droplets of varying sizes in the cells.Lipid droplet content in the silence and drug intervention groups was significantly reduced compared to the model group.Compared with the model group,the expression of IL-17 protein and mRNA were significantly decreased in the silence group,the low/medium/high-dose QZYGD groups and the polyene phosphatidylcho-line group(P<0.01),with the high-dose QZYGD group showing a more significant reduction.Conclusion QZYGD may exert protec-tive effects on NASH cells by inhibiting the expression of the inflammatory factor IL-17,alleviating inflammatory responses,and ameli-ating lipid metabolism.关键词
