基于PI3K/AKT/PTEN信号通路探讨补中益气汤调控Th17细胞免疫代谢改善AIT的作用机制
Exploring the mechanism of Buzhong Yiqi Decoction(补中益气汤)in relieving autoimmune thyroiditis by regulating Th17 immunometabolism based on the PI3K/AKT/PTEN signaling pathway

Abstract
Objective To explore the mechanism of Buzhong Yiqi Decoction(补中益气汤,BZYQD)in regulating abnormal immune me-tabolism of Th17 cells to relieve autoimmune thyroiditis(AIT)based on the PI3K/AKT/PTEN signaling pathway.Methods 40 NOD.H-2h4 mice were randomly divided into the blank group,the model group,the BZYQD group(9.56 g·kg-1)and the Western medicine group(selenium yeast tablet,3.033×10-5 g·kg-1).The blank group was gavaged with distilled water,and the remaining groups were gavaged with 0.05%aqueous sodium iodide.Each drug administration group was gavaged with liquid medication,while the blank group and the model group were gavaged with equal volume distilled water for 8 weeks.Histopathological changes and lymphocyte infiltration in thyroid gland were observed by HE,serum TGAb and TPOAb were detected by ELISA,PI3K,p-PI3K,AKT,p-AKT,PTEN,GLUT1,PKM2,HK2,and LDHA by Western blot(WB),and IL-17 and RORyt and the localized expression levels of IL-17 and RORγt in thyroid tissues was measured by immunohistochemistry(IHC).Results Compared with the blank group,the model group ex-hibited significant lymphocyte infiltration in the thyroid tissues under light microscopy,a marked increase in serum TGAb and TPOAb levels(P<0.01),a significant elevation in protein expression of p-PI3K/PI3K,p-AKT/AKT,GLUT1,HK2,PKM2,and LDHA in the thyroid tissues(P<0.01),and a notable decrease in PTEN protein expression(P<0.01).Additionally,the localization expres-sion of RORγt and IL-17 was significantly increased.Compared with the model group,all treatment groups showed significantly reduced lymphocyte infiltration in the thyroid tissues,a significant decrease in serum TGAb and TPOAb levels(P<0.01),a significant reduc-tion in protein expression of p-PI3K/PI3K,p-AKT/AKT,GLUT1,HK2,PKM2,and LDHA in the thyroid tissues(P<0.05,P<0.01),and a significant increase in PTEN protein expression(P<0.01).Furthermore,the localization expression of RORyt and IL-17 was significantly decreased.Conclusion BZYQD can ameliorate the imnmunometabolic abnormality of Th17 cells in AIT,possibly through the PI3K/AKT/PTEN-glycolysis axis as its intervention mechanism.关键词
