茯苓杏仁甘草汤调控脂肪酸β氧化对动脉粥样硬化斑块稳定性的影响
Effect of Fuling Xingren Gancao Decoction(茯苓杏仁甘草汤)on atherosclerotic plaque stabil-ity by regulating fatty acid β-oxidation

Abstract
Objective To investigate the mechanism of Fuling(Sclerotium Poriae Cocos)Xingren(Semen Armeniacae Amarum)Gan-cao(Radix et Rhizoma Glycyrrhizae)Decoction(茯苓杏仁甘草汤,FXGD)in enhancing the stability of atherosclerotic(AS)plaques.Methods A ApoE-/-mouse AS model and a smooth muscle cell lipid accumulation model were established.Histopathological changes in the mouse tissues were observed using hematoxylin-eosin(HE)staining,Sirius Red staining,and immunofluorescence.Lipid depo-sition in mice and cells was assessed by Oil Red O staining.RT-PCR and Western blot(WB)were employed to measure the expression levels of peroxisome proliferator-activated receptor α(PPARα)and fatty acidβ-oxidation-related factors of CPT1,CPT2,Acox1,Acox3 in the mouse tissues and cells.Results FXGD increased the content of α-smooth muscle actin(αSMA)and collagen while re-ducing matrix metalloproteinase-2(MMP2)expression in AS plaques.Additionally,FXGD upregulated PPARα and fatty acid β-oxi-dation-related factors of CPT1,CPT2,Acox1,Acox3 in the liver tissues of AS mice and in lipid-loaded smooth muscle cells in the model group.Furthermore,it counteracted the inhibitory effects of the PPARα inhibitor Norathyriol on PPARα and its downstream β-oxidation-related factors.Conclusion FXGD enhances AS plaque stability by activating the PPARα signaling pathway and promoting fatty acid[3-oxidation,thereby reducing lipid accumulation.关键词
