鹿角胶-酸枣仁调控PI3K/AKT/mTOR信号通路对CUMS大鼠神经保护的机制研究
A mechanism study on the neuroprotective effects of Colla Cornus Cervi-Semen Ziziphi Spi-nosae viaregulating the PI3K/AKT/mTOR signaling pathway in CUMS rats

Abstract
Objective To investigate the mechanism by which Colla Cornus Cervi-Semen Ziziphi Spinosae(鹿胶-酸枣仁,CCC-SZS)regulates the PI3K/AKT/mTOR pathway to ameliorate hippocampal neuronal damage in a CUMS depression rat model.Methods SPF-grade male SD rats were selected for modeling using chronic unpredictable mild stress(CUMS).After adaptive feeding,rats were treated with different concentrations of CCC-SZSvia gavage.Depression of rats was assessed using the forced swimming test,open field test,and sucrose preference test.Cell morphology was observed through HE staining.Immunohistochemistry(IHC)was used to detect the positive expression of BDNF and NGF proteins in the hippocampus.Enzyme-linked immunosorbent assay(ELISA)were conducted to measure serum contents of IL-6,IL-1β,TNF-α,and 5-HT,and Western blot(WB)was performed to detect the expression of signal pathway-related proteins in the hippocampus.Results Compared with the model group,all treatment groups showed improved behavioral indices in varying degrees.HE staining showed improved neuronal structure,reduced loss of neuronal cell damage and de-creased inflammatory cell infiltration.Serum levels of IL-6,IL-1β,TNF-α were significantly decreased(P<0.01,P<0.05),while 5-HT levels were increased(P<0.01,P<0.05).CCC-SZSupregulated the phosphorylation levels of signaling pathway-relat-ed proteins in the hippocampus(P<0.01,P<0.05).IHC indicated increased protein expressions of NGF and BDNF in the hippocam-pus(P<0.01).Conclusion CCC-SZSmay protect hippocampal neurons in CUMS rats by upregulating the PI3K/Akt/mTOR signaling pathway to modulate neurotransmitter and inflammatory levels.关键词
