基于铁死亡对葛花作用于MAPK3干预肝癌的机制研究
Research on the mechanism of flos puerariae intervening in liver cancer by acting on MAPK3 based on ferroptosis

Abstract
To investigate the effect and mechanism of flos puerariae in intervening in liver cancer by inducing ferroptosis,the databases such as GeneCards were utilized to screen liver cancer targets,ferroptosis targets were obtained from the FerrDb database,andflos puerariae targets were acquired from the TCMSP database.The intersection targets among liver cancer,fer-roptosis,and flos puerariae were identified using R language.With the assistance of String database and Cytoscape 3.9.1 soft-ware,a protein-protein interaction network(PPI)was constructed and screened for Hub genes using the CytoHubba plugin and R language.The David database and Microbioinfo were used for GO and KEGG analysis of the key action targets.To further screen the key targets,differential expression analysis,protein atlas analysis,clinical correlation analysis,and survival anal-ysis were performed on the Hub genes.The screened Hub genes were further validated through molecular docking and molec-ular dynamics simulation,and in vitro experimental validation was conducted using Western blot.A total of 5 859 liver cancer disease targets,485 ferroptosis genes,and 526flos puerariae targets were obtained,with 66 intersection targets.Seventeen Hub genes were screened out.By GO functional analysis the top 5 biological processes,cellular components,and molecular functions with the smallest P-values were selected;By KEGG enrichment analysis multiple pathways were identified.Differ-ential expression analysis,clinical correlation analysis,and survival analysis results indicated that the expression of SR C and MAPK3 genes in liver cancer was significantly higher than that of the normal group.Molecular docking and molecular dynam-ics simulation analysis results showed that MAPK3 had a good binding affinity with genistein,which is an active component of flos puerariae.Western blot experiments demonstrated thatflos puerariae significantly reduced the expression of P-MAPK3(P<0.05).In conclusion,flos puerariae can intervene in liver cancer by regulating ferroptosis through its action on MAPK3.关键词
