Abstract

Objective:To explore the role of Interleukin-32(IL-32)in the development of cervical cancer and the related molecular mechanism. Methods:Gene interference and construction of overexpression plasmid were used for knockdown and overexpression of IL-32 level in cells of cervical cancer.Cell proliferation activity was detected by CCK-8 assay,cell migration was detected by scratch healing assay,cell invasion was detected by Transwell assay,gene expression levels of IL-32,miR-141-5p and platelet-derived growth receptor alpha(PDGFRA)in cells were detected by quantitative real time polymerase chain reaction(qRT-PCR)assay,and expression levels of IL-32 protein in tissues were detected by immunohistochemical assay.Bioinformatics methods predicted the target gene relationship between miR-141-5p and PDGFRA. Results:IL-32 was significantly increased in tissues of cervical squamous cell carcinoma and cell lines of cervical cancer(SiHa and HeLa)when compared with those of paracancer tissues and normal cervical epithelial cell line HCerEpiC(all P<0.05).In cervical cancer cell line SiHa,IL-32 knockdown significantly inhibited cell proliferation,migration and invasion(P<0.05).In SiHa cell,knockdown or overexpression of IL-32 significantly decreased or increased the expression of miR-141-5p(P<0.05).Knockdown of miR-141-5p also significantly inhibited the proliferation,migration and invasion of SiHa cell(P<0.05).Bioinformatics methods predicted that PDGFRA was the target gene of miR-141-5p.Both IL-32 and miR-141-5p could reverse regulate the expression of PDGFRA(P<0.05). Conclusions:IL-32 is highly expressed in cervical squamous cell carcinoma,which may improve the proliferation,migration and invasion of cancer cells through miR-141-5p/PDGFRA,and promotes the occurrence and development of cervical squamous cell carcinoma.
