Abstract

To explore the biological function of foxtail millet SiDIR1 involved in drought resistance and salt tolerance,a foxtail millet nsLTP family member,SiDIR1,was cloned using 'Yugu 1' as the material,and an overexpression vector pCAM-SiDIR1 was constructed.The transgenic lines of Nicotiana benthamiana overexpressing SiDIR1 were obtained through Agrobacterium-mediated genetic transformation method,and the changes of morphological and physiological indices of wild-type(WT)and SiDIR1 transgenic plants were analyzed under the treatment of normal condition(sterile water,CK),drought(0.033 mol/L PEG-6000)and salt(0.200 mol/L NaCl)stress.The results showed that:Under drought and salt stress conditions,overexpression of millet SiDIR1 gene in tobacco plants could significantly decrease the leaf wilting index,and significantly increase the leaf surface area and above-ground fresh weight,indicating that the introduction of millet SiDIR1 gene into tobacco plants could enhance the drought resistance and salt tolerance of plants;overexpression of SiDIR1 could significantly increase the relative content of chlorophyll(SPAD)in tobacco leaves;overexpression of SiDIR1 could significantly increase the activities of peroxidase(POD)and superoxide dismutase(SOD),and significantly decrease the content of malondialdehyde(MDA)in tobacco,thereby reducing the accumulation of reactive oxygen species(ROS)and the damage of cell membrane.In conclusion,the heterologous expression of SiDIR1 in N.benthamiana could effectively improve the drought and salt tolerance of plants,and SiDIR1 might improve the drought resistance and salt tolerance of plants by maintaining the stability of chlorophyll,activating the plant oxidative defense and alleviating the cellular oxidative damage.
