Abstract

To screen the ARF genes related to anthocyanin synthesis in colored potatoes,based on the transcriptome sequencing data of potatoes,bioinformatics methods were used to analyze the physicochemical properties,phylogenetic evolution,conserved motifs,and cis-acting elements of the promoter of the gene family members.The expression patterns of nine StARFs genes related to anthocyanin synthesis in different growth stages of purple-flesh and white-flesh potatoes were verified by qRT-PCR.The results showed that:A total of 55 StARFs genes were screened and divided into four subfamilies.Members of the same subfamily have similar conserved motifs and domains,which indicate their similar functions.The results of the prediction of the cis-elements of the promoters showed that,nine StARFs contained hormone response elements,and five StARFs contained MYB binding site response elements,which might cooperate with MYB genes to regulate anthocyanin synthesis.The qRT-PCR results showed that seven StARFs among the nine StARFs were highly expressed in the yellow-skin white-flesh potato 'Atlantic' and lowly expressed in 'Huasong 66';the other two StARFs were highly expressed in the purple-skin purple-flesh potato 'Huasong 66' and lowly expressed in 'Atlantic'.The nine StARFs genes were highly likely to be involved in the biosynthesis process of anthocyanin in potato tubers.
