枳幼苗实时光合参数测定方法改进
Improvement of the method for determination of real-time photosynthetic parameters of trifoliate orange seedlings

Abstract
When a portable photosynthesis system(photosynthesizer)with a transparent bottom leaf chamber(a common method)is used to determine the real-time photosynthetic parameters of trifoliate orange seedlings,the leaves of trifoliate orange cannot completely cover the leaf chamber,and cumbersome methods such as paper-cutting and weighing are required to determine the actual working leaf area inside of the leaf chamber.The petioles of trifoliate orange leave are short and not easy to rotate the transparent bottom leaf chamber to allow the leaves inside of the chamber vertically irradiated by natural light,which leads to a large variation in light intensity.In order to determine the real-time photosynthetic parameters of trifoliate orange seedlings more conveniently,the leaf chamber of red and blue light source was used to replace the transparent bottom leaf chamber.The light intensity of growing environment of trifoliate orange seedlings determined by the plant light analyzer was set as the light intensity of the leaf chamber of red and blue light source to replace the natural light.The leaf area of trifoliate orange seedlings inside the leaf chamber was measured by the Image J method.Determination of leaf area with Image J method is easy to operate and highly accurate,it can replace commonly used leaf area determination methods such as the paper-cut weighing method.The light intensity measured(set)by the plant light analyzer was significantly higher than that of the transparent bottom leaf chamber method,but the measured real-time photosynthetic parameters of trifoliate orange seedlings,such as net photosynthetic rate(Pn),stomatal conductance(Gs),transpiration rate(Tr),and intercellular CO2 concentration(Ci),were not significantly different from those of the commonly used methods,which indicated that the improved method could replace the commonly used methods.The total time for determining the real-time photosynthetic parameters of single trifoliate orange seedlings by the improved method was about 80%of that by the conventional method,which improved the work efficiency and saved the cost for purchase complete sets of special photosynthetic apparatus.The results could provide reference for the determination of real-time photosynthetic parameters of other plants bearing small size of leaves.关键词
