新疆加工番茄SlWRKY35基因的克隆及功能初步分析
Cloning and Functional Analysis of SlWRKY35 Gene from Processing Tomato in Xinjiang

Abstract
To preliminarily investigate the biological role of the SlWRKY35 gene in processing tomatoes,we cloned the gene by PCR and using the processing tomato 'Yaxin 87-5'as the material,analyzed the SlWRKY35 gene sequence by bioinformatics.The SlWRKY35 overexpression vector was constructed and transformed into processing tomatoes.The transgenic processing tomato plants were identified by PCR.In order to explore the preliminary function of SlWRKY35 gene,the chlorophyll content and chlorophyll precursor content of transgenic processing tomato leaves were determined,and the expression of genes related to chlorophyll synthesis pathway was analyzed.Based on the findings,the SlWRKY35 gene's open reading frame measured 1 146 bp and encoded 381 amino acids.The SlWRKY35 protein contains the WRKY structural domain and is an unstable protein.Chlorophyll content in processing tomato leaves overexpressing SlWRKY35 was significantly higher than the wild type,as were the content of chlorophyll precursor substances,and the expression of genes related to the chlorophyll synthesis pathway,including ALAD,CHLH,CHLM,POR,and CAO,was up-regulated.By up-regulating the expression of genes linked to the chlorophyll synthesis pathway,overexpression of SlWRKY35 increased the content of chlorophyll precursors,which in turn boosted the accumulation of chlorophyll in processing tomato leaves.This study provides a basis for further functional mechanism of tomato SlWRKY35 gene.关键词
