Abstract

Objective:To investigate the therapeutic effect of Jinzhi on lipopolysaccharide(LPS)-induced acute lung injury(ALI)in mice.Methods:Mice were divided into a blank control group(30 mice)and a model estab-lishment group(60 mice)using a random number table method.ALI was induced by intraperitoneal injection of LPS(dissolved in 0.9%sodium chloride solution,15 mg/kg).After successful modeling,mice were randomly as-signed to a model group and a Jinzhi treatment group,with 30 mice in each group.The Jinzhi group received oral administration of Jinzhi liquid(0.1 mL/10 g)every 12 hours,and samples were collected at three time points(12,24,36 h).No intervention was applied to the model and blank groups,and samples were collected uniformly at cor-responding time points.Levels of tumor necrosis factor(TNF)-α and interleukin(IL)-6 in bronchoalveolar lavage fluid(BALF)were measured by ELISA.Pathological changes in lung tissue were observed by HE staining,and apoptosis of alveolar epithelial cells was detected by TUNEL staining.Results:Compared with the blank group,levels of TNF-α and IL-6 in BALF were significantly increased at all time points in the model group(P<0.05),along with varying degrees of thickening of alveolar septa,inflammatory cell infiltration,and interstitial edema;the apoptosis rate of cells also significantly increased(P<0.05).Compared with the model group,levels of TNF-α and IL-6 in BALF were significantly decreased at all time points in the Jinzhi group(P<0.05),with reduced inflamma-tory cell infiltration and interstitial edema,and better fusion of ruptured alveoli;the apoptosis rate of cells was sig-nificantly lower(P<0.05).Conclusion:Jinzhi may alleviate sepsis-induced ALI by downregulating TNF-α and IL-6 levels and reducing apoptosis of alveolar epithelial cells.
