基于p38 MAPK/NF-κB炎症通路观察四子定喘汤对过敏性哮喘大鼠炎症反应的影响
Exploring the Effect of Sizi Dingchuan Decoction on Inflammatory Response in Allergic Asthmatic Rats Based on the p38 MAPK/NF-κB Inflammatory Pathway

Abstract
Objective:To investigate the therapeutic effect of Sizi Dingchuan Decoction on allergic asthmatic rats and its impact on the p38 MAPK/NF-κB inflammatory pathway.Methods:24 SD rats were randomly divided into four groups:a normal control group,a model group,a dexamethasone group,and a Sizi Dingchuan Decoction group,six rats in each group.An allergic asthma rat model was established using ovalbumin(OVA)induction.Af-ter modeling,the intervention groups received drug treatments.Lung function tests measured minute ventilation(MV)and tidal volume(TV);HE staining was used to observe pathological changes in lung tissue;modified Giem-sa staining was employed to observe mast cell counts;ELISA was used to measure serum levels of interleukin-4(IL-4),interleukin-13(IL-13),and immunoglobulin E(IgE);Western blotting detected the expression levels of phosphorylated p38(p-p38)and NF-κB proteins.Results:Compared with the normal group,MV and TV signifi-cantly decreased in the model group(P<0.01).HE staining showed extensive infiltration of inflammatory cells and thickening of alveolar septa;mast cell counts increased(P<0.01);serum IgE,IL-4,and IL-13 levels significantly increased(P<0.01);and the expression levels of p-p38 MAPK and NF-κB proteins in lung tissue were signifi-cantly increased(P<0.01).Compared with the model group,Sizi Dingchuan Decoction effectively improved lung function in OVA-induced asthmatic rats(P<0.01 or P<0.05),alleviated lung tissue lesions,inhibited inflammato-ry cell infiltration(P<0.05),reduced serum levels of IL-4,IL-13,and IgE(P<0.01),and downregulated the ex-pression of p-p38 MAPK and NF-κB proteins in lung tissue(P<0.01 or P<0.05).Conclusion:Sizi Dingchuan Decoction can effectively improve airway inflammation in asthmatic rats and reduce the expression of inflammatory factors,which may be related to the regulation of the p38 MAPK/NF-κB inflammatory pathway.关键词
