壮筋续骨汤调节骨骼生长因子IGF2对发育性髋关节脱位幼鼠骨生物力学及血液流变学的影响
The Effect of Zhuangjin Xugu Decoction on Bone Biomechanics and Hemorheology in Young Rats with Developmental Hip Dislocation through Bone Growth Factor IGF2

Abstract
Objective:To investigate the relationship between Zhuangjin Xugu Decoction and bone growth factor(IGF2),as well as its effects on bone biomechanics and hemorheology in young rats with developmental hip dislo-cation.Methods:27 Wistar young rats were randomly divided into three groups(n=9):the control group,model group,and treatment group.After 14 days of intragastric administration of normal saline or Zhuangjin Xugu Decoc-tion,blood samples were collected to detect changes in hemorheological indicators.After euthanasia by cervical dis-location,bone tissues were collected for bone biomechanical testing.Surrounding soft tissues were isolated from the femoral tissue samples of each group,and the expression levels of IGF2,alkaline phosphatase(ALP),osteopon-tin(OPN),as well as the phosphorylation levels of phosphatidylinositol-3-kinase(PI3K)and protein kinase B(Akt)were analyzed using reverse transcription PCR and Western blotting.Results:Under the influence of Zhuangjin Xugu Decoction,the expression level of IGF2 in the surrounding tissues of the femur significantly in-creased,along with elevated levels of the osteogenic markers ALP and OPN.The phosphorylation levels of PI3K and Akt also increased.Meanwhile,the biomechanical properties of the femur improved,and hemorheological indi-cators approached those of normal young rats.Conclusion:Zhuangjin Xugu Decoction can promote IGF2 expres-sion to improve bone development,positively affecting bone biomechanics and hemorheological changes in young rats with developmental hip dislocation.关键词
