麝香化瘀醒脑颗粒治疗脑出血急性期作用机制的蛋白组学研究
A Proteomics Study on the Mechanism of Shexiang Huayu Xingnao Granules in the Treatment of Acute Stage of Intracerebral Hemorrhage

Abstract
Objective:To observe the clinical efficacy of Shexiang Huayu Xingnao Granules in treating intrace-rebral hemorrhage(ICH)and to explore its possible mechanism from a proteomics perspective.Methods:A total of 141 patients were digitally randomized into a treatment group(TD)and a control group(CD)at a ratio of 2∶1,and the clinical efficacy was observed.Then,5 cases were randomly selected from each group(marked as TD1/7 and CD1/7 before and after treatment respectively)for proteomics studies,and a total of 10 samples were ob-tained.The samples were subjected to proteomic analysis by protein extraction,trypsin digestion,TMT labeling,High pH RP-HPLC classification,liquid chromatography-mass spectrometry detection,and bioinformatics analysis.GO enrichment analysis and KEGG pathway enrichment analysis were performed on differential proteins.Results:The curative effect of Shexiang Huayu Xingnao Granules combined with basic treatment was better than that of the control group.There were 46 differential proteins in the treatment group before and after treatment,including 18 up-regulated proteins and 28 down-regulated proteins.There were 32 total differential proteins in the control group before and after treatment,13 up-regulated and 19 down-regulated.GO enrichment analysis showed that be-fore and after treatment in the treatment group,there were 7 subclasses of biological processes,4 subclasses of cel-lular components,and 8 subclasses of molecular functions in the 19 most significantly enriched subclasses.Before and after treatment in the control group,there were 3 subcategories of biological processes,3 subcategories of cellu-lar components,and 4 subcategories of molecular functions in the 10 most significantly enriched subcategories.The results of KEGG enrichment showed that the enrichment of the PI3K-Akt signaling pathway,focal adhesion,and ECM-receptor interaction pathway was the most significant in the treatment group before and after treatment.The control group had the most significant enrichment in proteasome,cholesterol metabolism,and Staphylococcus aureus infection before and after treatment.Conclusion:Shexiang Huayu Xingnao Granules in the treatment of ICH may regulate the expression of Ang,BDNF,PDGF,etc.,promote angiogenesis,reduce pathological BBB perme-ability,regulate PI3K-Akt signaling pathway,focal adhesion and ECM-receptor interaction pathway to regulate the pathophysiological process in patients with ICH,and play a neuroprotective role.关键词
