Abstract

Objective:To explore the pathogenesis of SARS-CoV-2 infection combined with allergic diseases and estimated Chinese medicine by reverse network pharmacology and molecular docking.Methods:Common targets of the two diseases were obtained,and the protein-protein interaction network was constructed.Degree values were calculated to determine key targets,followed by GO and KEGG enrichment analysis.Key targets were then used to reversely collect Chinese medicine ingredients.High-degree value targets were selected for molecular docking validation with active ingredients.Corresponding Chinese medicine was identified from active ingredients,and the key target-active ingredient-Chinese medicine network was constructed.Chinese medicine with high-degree values was selected and categorized by four properties,five flavors,meridians and effects.Results:A total of 247 common targets were obtained,with 39 key targets having corresponding Chinese medicine ingredients information in the TCMSP database.After screening,there were 22 key targets matched with 41 significant Chinese medicine ingredients.Molecular docking validation was performed on the top 5 targets and Chinese medicine ingredients,showing stable and active combinations.GO enrichment analysis indicated biological processes such as inflammatory response,positive regulation of cytokine production,and innate immune response.KEGG enrichment analysis showed pathways such as SARS-CoV-2,Th17 cell differentiation,cytokine-cytokine receptor interaction,HIF-1 signaling pathway,and NF-κB signaling pathway.Cold and bitter medicines were the most common,followed by warm and sweet medicines,primarily targeting liver and lung meridians,mainly including heat-clearing,exterior-releasing,and tonifying medicines.Commonly used Chinese medicine included Kushen(Radix Sophorae Fiavescentis),Huangqin(Radix Scutellariae),Huangqi(Radix Astragali),Baishao(Radix Paeoniae Alba),and Renshen(Radix Ginseng)Conclusion:On the basis of existing research,this study identified the targets,signaling pathways,active ingredients and predicted Chinese medicine four properties,five flavors,meridians and effects,which can provide new ideas for clinical syndrome differentiation and treatment of these diseases.
