基于Mincle/Syk/NF-κB信号通路探究甘草泻心衍生方治疗IgA肾病大鼠作用机制
Exploration on the Mechanism of Gancao Xiexin Derivative Prescription in the Treatment of IgA Nephropathy Rats Based on Mincle/Syk/NF-κB Signaling Pathway

Abstract
Objective To explore the mechanism of Gancao Xiexin Derivative Prescription for the treatment of IgA nephropathy rats based on Mincle/Syk/NF-κB signaling pathway.Methods The rat model of IgA nephropathy was prepared by bovine serum albumin gavage+subcutaneous injection of carbon tetrachloride+LPS tail vein injection.The modeled rats were randomly divided into model group,losartan group and Gancao Xiexin group,with 10 rats in each group,and 9 rats were set as control group,.After successful modeling,losartan solution was given in losartan group and Gancao Xiexin Derivative Prescription solution was given in Gancao Xiexin group for eight weeks.After the intervention,urine red blood cell count,24 hUTP,SCr and BUN were detected by biochemical analysis;immunofluorescence staining was used to observe IgA deposition in renal tissue,HE staining,PAS staining and Masson staining were used to observe the morphology and fibrosis of renal tissue;the expressions of Mincle,Syk,NF-κBp65,IL-6,IL-1β,IP-10 and MCP-1 proteins in renal tissue were detected by Western blot.Results Compared with the control group,the model group rats had withered hair,reduced food and water intake,poor mental state,decreased body mass,significantly increased urine red blood cell count and 24 hUTP(P<0.01),the contents of SCr and BUN significantly increased(P<0.05),with significant increase in IgA deposition in renal tissue(P<0.01),structural disorder of glomeruli,dilation of renal tubules,significant infiltration of inflammatory cells,proliferation of mesangial cells,and increased deposition of collagen fibers;the protein expressions of Mincle,Syk,NF-κBp65,IL-6,IL-1β,IP-10 and MCP-1 in renal tissue significantly increased(P<0.01).Compared with the model group,the losartan group and Gancao Xiexin group showed improved mental state,food and water intake,increased body mass,significantly reduced urine red blood cell count and 24 hUTP(P<0.05,P<0.01),and the contents of SCr and BUN decreased,IgA deposition in renal tissue significantly decreased(P<0.05),glomerular and tubular structures improved,edema and inflammatory cell infiltration decreased,and collagen fiber deposition decreased;the protein expressions of Mincle,Syk,NF-κBp65,IL-6,IL-1β,IP-10 and MCP-1 in renal tissue were significantly decreased(P<0.05,P<0.01).Conclusion Gancao Xiexin Derivative Prescription can improve the immune barrier and inhibit the release of inflammatory factors by regulating the Mincle/Syk/NF-κB signaling pathway,thus delaying the occurrence and development of IgA nephropathy.关键词
