Abstract
Objective:Based on the theory of Qihua(气化)-autophagy,through network pharmacology and molecular docking methods,the mechanism of Huangqi(Astragali Radix)in treating different diseases type 2 diabetes and hypothyroidism was explored.Methods:The active ingredients of Huangqi were obtained from TCMSP database and literature search.The TCMSP,PubChem,SwissTargetPrediction,and PharmMapper databases were used to obtain relevant targets.The GeneCards,OMIM,and PharmGKB databases were used to predict the relevant targets of type 2 diabetes and hypothyroidism.A protein interaction(PPI)network of intersection targets was constructed by STRING database to predict core targets through network topology parameters,R language was used to perform core targets gene ontology(GO)and Kyoto encyclopedia of genes and genomes(KEGG)pathways enrichment analyses,and finally molecular docking and literature data were used to verify the binding ability of core components and core targets.Results:A total of 21 active compounds of Huangqi were obtained.The main active ingredients included quercetin,kaempferol,astragalus pteranoside,7-O-methylisomyxoside,isorhamnetin,arranol,etc.There were 110 intersection targets of Huangqi with type 2 diabetes and hypothyroidism,and 14 core targets including AKT1,PPARG,TNF,IL-6,and TP53 were obtained through two screenings.GO and KEGG analyses showed that the core target mainly exertsed regulatory effects through multiple signaling pathways such as PI3K-Akt,MAPK,and AGE-RAGE signaling pathway.The core component and core target molecular docking showed good binding activity.Conclusion:Type 2 diabetes and hypothyroidism are related to each other through the"Qihua-autophagy"theory.Huangqi can treat type 2 diabetes and hypothyroidism by regulating"Qihua-autophagy"with multi-components,multi-targets,and multi-pathways.关键词
