枸杞红瘿蚊成虫触角转录组及嗅觉相关基因分析
Antennal Transcriptome and Olfaction-related Genes of Gephyraulus lycantha(Diptera:Cecidomyiidae)

Abstract
Objective:To obtain the antennal transcriptome data of Gephyraulus lycantha Jiao&Kolesik and mine the olfaction-related genes.Methods:The Illumina NovaSeq 6000 platform was used for transcriptome sequencing of the antennae of male and female adults of G.lycantha,and gene annotation was performed across multiple public databases,including Non-Redundant Protein Database(NR),Nucleotide Sequence Database(NT),Protein Families Database(Pfam),Non-Redundant Protein Database(KOG)/Clusters of Orthologous Groups of proteins(COG),Swiss-Prot,Kyoto Encyclopedia of Genes and Genomes(KEGG)Orthologue(KO),and Gene Ontology(GO).Bioinformatics and phylogenetic analyses were then carried out for the olfaction-related genes screened out.The differentially expressed genes in male and female antennae were validated by using quantitative real-time PCR(qRT-PCR).Results:A total of 54 105 unigenes were obtained,with an average length of 1 102 bp and a N50 of 1 841 bp.Among them,19 013 unigenes were successfully annotated in at least one database.Functional annotation identified 152 olfaction-related genes,including 27 odorant-binding proteins(OBPs),3 chemosensory proteins(CSPs),86 odorant receptors(ORs),15 gustatory receptors(GRs),15 ionotropic receptors(IRs),and 6 sensory neuron membrane proteins(SNMPs).Phylogenetic analysis revealed three GlycOBPs clustered in the OS-E/OS-F clade.GlycCSPs exhibited high bootstrap support with other dipteran homologs.Sixteen GlycORs(GlycOR3/7/9/11/14/23/26/44/53/60/61/67/71/80/83/85)formed a distinct species-specific clade.Both GlycIRs and GlycGRs showed strong phylogenetic congruence with orthologs from other dipteran species.Notably,GlycSNMP1 and GlycSNMP2 were clustered into the conserved SNMP1 and SNMP2 clades,respectively,with high bootstrap support among dipteran species.The qRT-PCR results demonstrated six differentially expressed genes(encoding GlycOBP2,GlycOBP14,GlycOBP15,GlycOR3,GlycOR9,and GlycOR85)related to olfaction in male and female antennae,which were consistent with transcriptome sequencing results.Conclusion:The results significantly enrich the available information on the olfaction-related genes of G.lycantha and lay a molecular foundation for delving into the olfactory recognition mechanisms and controlling this pest by targeting olfaction-related genes.关键词
