摘要

Abstract

Objective:This study aims to investigate the effects of exogenous coenzyme Q10 on the growth of hairy roots of Arnebia euchroma,the content of total naphthoquinone and shikonin compounds,and the activity of antioxidant enzymes,thereby providing a reference for reducing the browning of the hairy roots and increasing shikonin content.Methods:Exogenous coenzyme Q10 at a concentration of 20 μmol·L-1 was added during the cultivation process of hairy roots of A.euchroma.The biomass,total naphthoquinone and shikonin compound contents,antioxidant enzyme activities,reactive oxygen species(ROS)levels,and malondialdehyde(MDA)content of hairy roots of A.euchroma were measured.In addition,transcriptome sequencing analysis was performed on the hairy root samples.Results:Compared with the control group,coenzyme Q10 treatment significantly increased the content of total naphthoquinone and shikonin compounds(P<0.05).Moreover,the activities of superoxide dismutase,catalase,and peroxidase were significantly enhanced(P<0.01),while ROS and MDA levels were significantly reduced(P<0.01).Transcriptome analysis identified a total of 561 differentially expressed genes,of which 154 were upregulated,and 407 were downregulated.Gene function enrichment analysis showed that these genes were mainly involved in defense responses,antioxidant responses,and redox pathways related to monooxygenase activity.Furthermore,the expression levels of antioxidant enzyme genes related to ROS scavenging were significantly upregulated.Conclusion:Exogenous coenzyme Q10 can regulate the antioxidant enzyme activity and related metabolic pathways in A.euchroma hairy roots,,significantly increase the content of shikonin compounds and effectively mitigate the browning of hairy roots.
