摘要

Abstract

Benzo[a]pyrene(BaP),an injurant produced in high-temperature processing foods,has been reported to cause colon injury.This study sought to investigate whether habitual consumption of whole grain buckwheat(WGB)or oat(WGO)could prevent 50 mg/kg BaP-induced colon injury of BALB/C mice.The results showed that both WGB and WGO delayed BaP-induced undesirable trend of body mass,food intake and water intake,as well as reversed high expression of metabolic enzymes,i.e.,aryl hydrocarbon receptor(AhR),cytochrome P450 family 1 subfamily A member 1(CYP1A1),CYP1B1 and glutathione S-transferase P1(GST-P1).WGB and WGO showed significant inhibitory effects on BaP-induced cell pyroptosis indexes,i.e.,NACHT,LRR and PYD domain-containing protein 3(NLRP3),apoptosis-associated speck-like protein containing a CARD(ASC),cysteinyl aspartate specific proteinase-1(Caspase-1),gasdermin-D(GSDMD),tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),IL-18,inducible nitric oxide synthase(iNOS)and cyclooxygenase-2(COX-2).Both WGB and WGO increased the abundance of Bacteroidetes and decreased the abundance of Firmicutes,Verrucomicrobia and Proteobacteria,thus improving the intestinal flora imbalance induced by BaP.By applying integration analysis,30 bacterial taxa and their 13 metabolic indicators were identified as key signatures.They synergistically promoted WGO and WGB to reduce BaP-induced colon injury,and WGB and WGO prevented BaP-induced colon injury mainly by regulating intestinal flora and short-chain fatty acids(SCFAs).This study offered a new theoretical foundation and dietary guidance for mitigating colon injury induced by harmful substances in food processing through the dietary supplementation of WGB and WGO.
