Tongdu Xingshen Electroacupuncture Alleviates Learning and Memory Impairment via Metabolic Pathway in Cerebral Ischemia-Reperfusion Rats

Abstract
Objective To explore the metabolomic changes and possible mechanism of Tongdu Xingshen electroacupuncture in alleviating middle cerebral artry occlusion/reperfusion(MCAO/R)learning and memory impairment rats.Methods A total of 90 SD rats were divided into a control group(n=15),sham group(n=15),and 60 MCAOR rat models were established by thread suppositing method.Totally 38 rats successfully modeled were randomly divided into a model group(n=19)and a Tongdu Xingshen electroacupuncture group(electroacupuncture group,n=19).In the electroacupuncture group,Shenting(GV24)and Baihui(GV20)acupoints were electroacupunctured for 30 minutes each time,with a frequency of 2/10 Hz and a current intensity of 1~3 mA,once a day for 14 consecutive days.No intervention was given to the other groups.The Zea-Longa scoring method and Morris water maze behavioral experiments were employed to assess the neurological function score and learning and memory function of the rats.HE staining was utilized to observe the injury of hippocampal neurons in rats,and non-targeted metabolomics technology was applied to detect metabolites in the hippocampus of rats and screen for differential metabolites and related metabolic pathways.Results Compared with the sham group,the model group showed a significant increase in neurological function scores(P<0.01),a significant decrease in learning and memory function(P<0.01),and significant damage to hippocampal neurons.Compared with the model group,the electroacupuncture group showed a significant decrease in neurological function scores(P<0.01),a significant improvement in learning and memory function(P<0.01),and a significant recovery in hippocampal neuronal damage.The metabolomics results indicated that acupuncture treatment with Tongdu Xingshen can significantly increase the levels of metabolite thiamine,beta-alanine,acetylcholine,and inosine and decrease the levels of 4-hydroxy-2-butenoic acid gamma-lactone,L-acetylcarnitine,butyrylcarnitine,2-methylbutyroylcarnitine and cytarabine in the hippocampal tissue of MCAO/R model rats.At the same time,it could affect thiamine metabolism,pantothenate and CoA biosynthesis,beta-alanine metabolism,propanoate metabolism,glycerophospholipid metabolism,pyrimidine metabolism and purine metabolism pathways in the hippocampus of rats.Conclusions Tongdu Xingshen electroacupuncture may ameliorate learning and memory deficits in MCAO/R model rats by modulating differential metabolites through key metabolic pathways(thiamine metabolism,beta-alanine metabolism,glycerophospholipid metabolism,pyrimidine metabolism).This intervention regulates metabolic disturbances in hippocampal tissue,mitigates hippocampal neuronal damage,and thereby enhances learning and memory function.关键词
