摘要

Abstract

Objective To observe the regulatory effect and mechanism of Tongdu Tiaoshen electroacupuncture on cerebral glucose metabolism in Alzheimer's disease(AD).Methods Seventy-five SD rats were randomly divided into the sham operation group,model group,acupuncture group,inhibitor group,and acupuncture+inhibitor group,with 15 rats in each group.AD model was established by injection of Aβ25-35 into lateral ventricle,with the acupuncture group receiving electroacupuncture treatment at Baihui(DU20)and Shenting(DU20)points,and the inhibitor group receiving lateral ventricle injection of phosphatidylinositide 3-kinases(PI3K)/protein kinase B(AKT)inhibitor LY294002.The acupuncture+inhibitor group received electroacupuncture treatment and LY294002 injection.Morris water maze was used to evaluate the cognitive ability of rats.Nissl staining was used to observe the pathological changes in the hippocampus.Immunohistochemistry was used to observe the positive expression of Aβ in hippocampus.18F-FDG-PET was used to observe the glucose metabolism in the brain.Western Blot was used to detect the expression of glucose transport and regulatory proteins[glucose transporter1(GLUT1),GLUT3,GLUT4]in the hippocampus.Western Blot and RT-PCR were used to detect protein and mRNA expression levels of insulin signals[insulin receptor(INSR),PI3K,p-AKT,glycogen synthase kinase-3β(p-GSK-3β)]in the hippocampus.Results Compared with the model group,the acupuncture group showed significant improvement in cognitive ability and hippocampal neuron damage,and a significant increase in brain glucose uptake rate(P<0.05).The positive expression of Aβ and proteins levels of GLUT1,GLUT3,and GLUT4 in the hippocampus were significantly increased(P<0.05).At the same time,the levels of INSR,PI3K,p-AKT,and p-GSK-3β in the hippocampus were significantly increased(P<0.05).The inhibitor group showed reduced cognitive ability,aggravated pathological damage in the hippocampus,and decreased glucose uptake rate in the brain(P<0.05).The expression of Aβ and the protiens expressions of GLUT1,GLUT3,and GLUT4 in hippocampal tissue decreased(P<0.05),while the protein and mRNA levels of INSR,PI3K,p-AKT,and p-GSK-3β decreased(P<0.05).Compared with the acupuncture group,the acupuncture+inhibitor group showed reduced cognitive ability,aggravated pathological damage in the hippocampus,and decreased glucose uptake rate in the brain(P<0.05).The expression of Aβ and the protiens expressions of GLUT1,GLUT3,and GLUT4 in hippocampal tissue decreased(P<0.05),while the protein and mRNA levels of INSR,PI3K,p-AKT,and p-GSK-3β decreased(P<0.05).Conclusions Tongdu Tiaoshen electroacupuncture can improve the glucose metabolism level in the brain of AD rats.Its mechanism is related to the activation of the central insulin PI3K/Akt/GSK-3β pathway.
