Curcumin Inhibits Proliferation and Migration in HepG2 Cells by Upexpression of PDCD4 and PXR

Abstract

Objective To investigate the effect of curcumin on the proliferation and migration in human hepatoma HepG2 cells,and explore the underlying molecular mechanism.Methods HepG2 cells were cultured in vitro and treated with different concentrations of curcumin(0,2.5,5,10,20,50 μmol/L)for 48 hours.The cell viability was detected by the cell morphology observation and cell counting kit-8(CCK-8)methods.The cell migration was observed by the cell scratch assay.HepG2 cells were pretreated with sulforaphane[SFN,a common suppressor of pregnane X receptor(PXR)]for 4 hours and then stimulated with curcumin.The mRNA and protein expressions of programmed cell death factor 4(PDCD4),PXR and target genes including multidrug resistance gene 1(MDR1)and cytochrome P450 3A4(CYP3A4)were detected by the qRT-PCR and Western Blot respectively.Results Compared with control group,the survival rate of HepG2 cells in 5,10,20 and,50 μmol/L curcumin groups decreased(P<0.001,P<0.01,P<0.05),the cell migration rate of the 2.5,5,10 μmol/L groups decreased after 72 hours(P<0.01,P<0.05).Compared with control group,the protein and mRNA expression of PDCD4 increased in the 20 and 50 μmol/L groups(P<0.01,P<0.05),PXR and its target genes MDR1 and CYP3A4 also significantly increased(P<0.001,P<0.01,P<0.05),SFN administration alleviated PDCD4 expression induced by curcumin(P<0.001).Conclusion Curcumin can inhibit the HepG2 cells proliferation and migration,and its mechanism might be related to upregulation of PDCD4 by activating PXR.
