Effect of Ginsenoside Rd on Ang Ⅱ Induced Cardiac Remodeling and Inflammation

Abstract
Objective To investigate the effect of ginsenoside Rd(GRd)on angiotensin Ⅱ(Ang Ⅱ)induced cardiac remodeling and inflammation.Methods Six-week-old male C57BL/6 mice were infused with Ang Ⅱ(1 μg/(kg·min))via subcutaneous implantation of a micro-osmotic pump without interruption every day for 4 weeks to establish a model of hypertensive cardiac hypertrophy and excessive remodeling.In contrast,mice in control group were infused with saline and mice in GRd group were given GRd by gavage during the last 2 weeks.24 mice were randomly divided into four groups:control group(CON),model group(Ang Ⅱ),low-dose group(Ang Ⅱ+25 mg/kg),and high-dose group(Ang Ⅱ+50 mg/kg),with six mice in each group.Blood pressure and body mass were measured weekly,and cardiac function was assessed by cardiac ultrasound at week 4.Serum and cardiac tissues were collected,and myocardial injury was assessed by enzyme-linked immunosorbent assay(ELISA)to determine serum levels of Ang Ⅱ,atrial natriuretic peptide(ANP),and creatine kinase-myocardial band(CKMB).Cardiac hypertrophy was evaluated using hematoxylin-eosin staining,while myocardial fibrosis was assessed using Masson's trichrome and Sirius red staining.RT-qPCR was performed to detect mRNA levels of cardiac tissue hypertrophy and fibrosis indicators(Myh7,Col1a1,and Tgfb1)as well as inflammatory genes(Il1b,Il6,and Tnf).Results The results of blood pressure measurements in mice showed that all Ang Ⅱ-infused mice exhibited significantly higher systolic blood pressure compared to control group.Additionally,the ELISA kit results indicated that the serum levels of Ang Ⅱ in the Ang Ⅱ-infused mice were all significantly higher(P<0.01).Compared with model group,the levels of ANP and CKMB in the serum of mice in administered group were significantly reduced(P<0.01).Hematoxylin-eosin staining of cardiac tissues demonstrated that GRd could effectively alleviate cardiac hypertrophy in mice.Furthermore,Masson's trichrome and Sirius red staining re-vealed a significant reduction in fibrosis of cardiac tissues.RT-qPCR analysis showed that GRd significantly alleviated Ang Ⅱ induced cardiac hypertrophy,fibrosis,and inflammation-related gene expression(P<0.05,P<0.01).Conclusion GRd attenuates Ang Ⅱ induced cardial hypertrophy,fibrosis,and inflammatory response.关键词
