神经干细胞来源外泌体对MPTP帕金森病小鼠模型的治疗作用
Therapeutic Effect of Exosomes Derived from Neural Stem Cells on MPTP Induced Parkinson's Disease Model Mice

Abstract
Objective To explore therapeutic effect and possible mechanism of exosomes(Exos)on 1-methyl-4-phenyl-1,2-3,6-tetrahydro pyridine(MPTP)induced Parkinson's disease(PD)model mice.Methods Exos was extracted by differential centrifugation and identified by transmission electron microscopy and Western blot.Mice were randomly divided into control group(NC),model group(MPTP),and Exos treatment group(EXO).The MPTP and EXO groups were injected intraperitoneally with MPTP,while Exos was injected into the tail vein of EXO group.Behavioral observations were conducted at the end of drug administration,and at the conclusion of behavioral test,blood was collected from eyeballs.The mice were euthanized by medullary dissection,and brain tissues were harvested.The levels of superoxide dismutase(SOD),glutathione(GSH),and malondialdehyde(MDA)in serum were measured,and the expression of antioxidant-related proteins in brain tissues was assessed using Western blot analysis.Results Transmission electron microscopy observations showed that the extracted Exos was in the shape of a bowl torus,which was consistent with the morphological characteristics of Exos.Western blot results indicated that the Exos marker proteins CD9 and CD63 were expressed.The results of biochemical indexes showed that,compared with NC group,the serum levels of SOD and GSH in the mice of MPTP group decreased significantly(P<0.01),while the content of MDA increased significantly(P<0.01).In comparison to MPTP group,the serum levels of SOD and GSH in the mice of EXO group increased(P<0.01),and the content of MDA decreased(P<0.05).Additionally,the results of Western blot analysis indicated that the protein expression of Nrf2 and HO-1 in the brain tissues of mice in MPTP group was significantly lower(P<0.01)compared to NC group.Conversely,the protein expression of Nrf2 and HO-1 in the brain tissues of mice in EXO group was significantly higher(P<0.01)compared to MPTP group.Conclusion Neural stem cell-derived Exos has a therapeutic effect on MPTP induced PD mice by a mechanism that may be related to antioxidant effect.关键词
