牛磺酸通过调节iNOS活性抑制心肌成纤维细胞增殖的作用机制
Mechanism of Taurine in Inhibiting Myofibroblasts Proliferation by Regulating iNOS Activity

Abstract
Objective To investigate the inhibition of angiotensin Ⅱ(Ang Ⅱ)induced proliferation of rat pups myofibroblasts(myoFbs)by taurine(Tau)and to explore possible mechanisms involved.Methods Myocardial fibrosis(MF)model was established by using Ang Ⅱ to induce the proliferation of neonatal rat pups myoFbs in vitro.MTT was used to detect myoFbs proliferation;a hydroxyproline kit was used to measure collagen content;and immunocytochemical staining,laser confocal microscopy,and Western blot were used to detect the expression of inducible nitric oxide synthase(iNOS)in the cytoplasm of myoFbs.Results Compared to Ang Ⅱ group,Tau(30,60,120 mmol/L)significantly inhibited Ang Ⅱ induced myoFbs proliferation and collagen synthesis,with differences that were statistically significant(P<0.05,P<0.01)and exhibited dose-dependency.Immunocyto-chemical staining and laser confocal microscopy,combined with an image analysis system,showed that Tau could inhibit myoFbs proliferation by increasing the expression of iNOS in the cytoplasm of myoFbs and promoting protein synthesis,with differences that were statistically significant(P<0.05,P<0.01)when compared to Ang Ⅱ group.The results of Western blot analysis indicated that Tau could increase the expression of iNOS in the cytoplasm of myoFbs.Conclusion Tau inhibits MF by increasing the expression of iNOS in the cytoplasm of myoFbs,promoting NO synthesis,and inhibiting the proliferation of myoFbs and collagen synthesis.关键词
