Expression of PAX6 in Gastric Cancer Tissues and Its Effect on Cell Activity
摘要
Abstract
Objective To investigate the expression status of paired box gene 6(PAX6)in gastric cancer tissues and its effect on cell activity.Methods 60 patients with gastric cancer were selected as study subjects,and gas-tric cancer tissues and corresponding paracancerous normal tissues were collected.RT-PCR and Western blot were used to detect the mRNA and protein expression levels of PAX6,respectively.Human gastric cancer cells MGC-803 were cultured in vitro and divided into blank control group(conventional culture without other treat-ments),empty vector transfection group(transfected with empty vector plasmid),and PAX6 over expression group(transfected with PAX6 over expression plasmid).The cell cycle distribution,cell proliferation and migration abili-ty,as well as the protein expression levels of ADAM10,MMP2,and MMP9 were detected in each group.Results The expression levels of PAX6 mRNA and protein in gastric cancer tissues were lower than those in normal tissues adjacent to the cancer(P＜0.01).There were statistically significant differences in cell cycle dis-tribution of each group(P＜0.001).The percentage of cells in the G0/G1,phase in PAX6 over expression group was higher than that in empty vector transfection group and blank control group,while the percentage of cells in the S and G2/M phases was lower than that in empty vector transfection group and blank control group(both P＜0.001).At 5 and 7 d post-transfection,the cell proliferation ability in PAX6 over expression group was significantly lower than that in empty vector transfection group and blank control group(P＜0.05).The cell scratch assay re-vealed that the migration rate of cells in PAX6 over expression group at 24 h after culture was significantly lower than that in empty vector transfection group and blank control group(P＜0.05).The relative expression levels of ADAM10,MMP2 and MMP9 proteins in the gastric cancer cells of PAX6 over expression group were lower than those in empty vector transfection group and blank control group(P＜0.05).Conclusion PAX6 expression was down-regulated in gastric cancer tissues.Over expression of PAX6 was able to inhibit the proliferation and migra-tion of gastric cancer cells,which might be related to the down-regulation of ADAM10,MMP2 and MMP9 protein expression.关键词
