m7G甲基转移酶基因GhTRM8a/82调控棉花株高发育的研究

Abstract
m7G-methyladenosine(m7G),as one of the ubiquitous tRNAs modifications from bacteria to humans,plays an important role in regulation of gene expression.The core complex of m7G methyltransferase consists of METTL1 and its cofactor WDR4,but the role of their homologous proteins in plant growth and development has been rarely reported.In this study,we identified GhTRM8a and GhTRM82,the members of core complex of m7G methyltransferase in upland cotton,and found that TRM8a and TRM82 were highly conserved during plant evolution.GhTRM8a and GhTRM82 were expressed in various tissues,and their encoded proteins were localized in the nucleus and can interact to form heterodimers.Virus-induced gene silencing(VIGS)of GhTRM8a and GhTRM82 expression resulted in shorter stems and significantly shorter cotton plants.Stem longitudinal paraffin sections showed that the silencing of GhTRM8a and GhTRM82 significantly shortened the length of stem cells,indicating that GhTRM8a/GhTRM82 regulated the development of cotton plant height by affecting stem cell elongation.The qRT-PCR experimental results demonstrated that GhTRM8a and GhTRM82 regulate cotton stem development by influencing the expression of genes associated with gibberellin(GA)biosynthesis,as well as auxin and strigolactone pathways.In summary,GhTRM8a and GhTRM82 interacted to form a protein complex,which modulated cotton stem development through regulating GA biosynthesis.This study provides mechanistic insights into the regulation of plant architecture development in cotton.关键词
