玉米组蛋白去乙酰化酶ZmHDT102与ZmSWI3B蛋白的互作研究
Study on the interaction between maize histone deacetylase ZmHDT102 and ZmSWI3B protein

Abstract
Histone deacetylase ZmHDT102 is one of the histone deacetylases associated with maize disease resistance,and its mechanism has not been clarified.It has been reported that histone deacetylase AtHD2C can interact with AtSWI3B protein in Arabidopsis thaliana,but it is not clear whether the homologous protein ZmHDT102 of HD2C interacts with ZmSWI3B in maize.In this study,the ZmSWI3B protein in maize was identified by bioinformatics methods,and the interaction between ZmHDT102 and ZmSWI3B was identified by protein-protein molecular docking technology,yeast two-hybrid(Y2H)technology and double luciferase complementation(LCA)technology.The results showed that the molecular docking model of ZmHDT102-ZmSWI3B showed that there may be physical interaction between the two proteins.Y2H results showed that the yeasts co-transformed with ZmHDT102 and ZmSWI3B grew normally on the two-deficient(-Leu/-Trp),three-deficient(-Leu/-Trp/-His)and four-deficient(-Leu/-Trp/-His/-Ade)medium,while the yeasts transformed with the control combination could not grow normally on the three-deficient and four-deficient medium,indicating that ZmHDT102 and ZmSWI3B could interact directly in yeast cells.LCA results showed that the tobacco leaves infiltrated with the combination of ZmHDT102 and ZmSWI3B showed obvious fluorescence,while the tobacco leaves infiltrated with the control combination didn't show fluorescence signals,indicating that ZmHDT102 and ZmSWI3B could interact directly in tobacco.The results showed that there was an interaction between ZmHDT102 and ZmSWI3B,which laid a foundation for elucidating the regulatory mechanism of ZmHDT102 in maize disease resistance.关键词
