基于转录组测序的侧柏EST—SSR引物开发与鉴定分析
Development and identification analysis of EST-SSR primers for Platycladus orientalis based on transcriptome sequencing

Abstract
In order to evaluate and utilize Platycladus orientalis germplasm resources,this study developed EST-SSR primers based on transcriptome sequencing technology using roots,stems and leaves of Platycladus orientalis,and used yellow-leaved seedlings and green-leaved seedlings of half-sibling offspring of yellow-leaved Platycladus orientalis cv.MC1 to assess the applicability and polymorphism of the primers in different Platycladus orientalis resources.The results showed that the number of SSR sites in the root organ transcriptome was larger than that in the stem and leaf organs.The SSR repeat types in leaf organs were larger than those in stem and root organs,and the dominant motifs in all three organs were A/T,AG/CT,and AAG/CTT.The number of SSRs with lengths of 10,15,and 18 bp was the highest in the three organs.60 pairs of EST-SSR primers had an efficiency rate of 100%and a polymorphism rate of 16.67%,and the variation of the PIC values of the different primers ranged from 0.322 to 0.722.Using the amplification product data of 10 pairs of polymorphic primers,the samples were subjected to cluster analysis and genetic analysis,and it was hypothesized that 50% of MC1 half-siblings belonged to the heterozygous offspring,and the rest were suspected to be the autogamous offspring,and that the population of heterozygous offspring and the population of suspected autogamous offspring were classified into 13 taxa at a genetic distance of 200.关键词
