Abstract

To elucidate the function of CsPIP2;4 in salt stress tolerance of cucumber,this study introduced the empty plasmid of the pCAMBIA2300s and the pCAMBIA2300s:CsPIP2;4 plasmid into Agrobacterium rhizogenes strain K599,respectively.Composite cucumber plants overexpressing CsPIP2;4(OP)and empty vector-transformed(EV)plants were generated via hypocotyl injection.Morphological characteristics and physiological indexes,including malondialdehyde(MDA)content,soluble sugar and protein levels,and superoxide dismutase(SOD)activity,were comparatively analyzed under salt stress.Results demonstrated that OP composite plants exhibited superior growth phenotypes compared to EV controls under salt stress,with reduced leaf chlorosis and marginal scorching symptoms,as well as more developed root systems.Concurrently,OP plants showed decreased MDA content and SOD activity in leaves,alongside elevated soluble protein and sugar levels.All measured parameters displayed significant or highly significant differences between OP and EV plants after stress treatment for 7 days.These findings indicated that CsPIP2;4 positively regulated salt tolerance of cucumber,which was closely associated with its promotion of osmotic adjustment substances accumulation,particularly soluble sugars and proteins.
