枯草芽孢杆菌菌株Z-14的抑菌活性及其对玉米秸秆青贮发酵的影响
The antimicrobial activity of Bacillus subtilis strain Z-14 and its influence on the silage fermentation of corn stover
摘要
Abstract
The aerobic decay of silage corn stalks after fermentation seriously affects their nutritional quality and produces a large amount of fungal toxins that affect animal and human health.Bacillus subtilis produces various secondary metabolites and has received widespread attention in silage feed research.This study detected the inhibitory activity of the fermentation supernatant of B.subtilis Z-14 on different types of pathogenic fungi and bacteria through perforation method.The ability of its synthetic digestive enzymes was detected by plate confrontation method and perforation method.Whole genome sequencing combined with analysis via the antiSMASH(Antibiotics&Secondary Metabolite Analysis Shell)database was conducted to identify gene clusters associated with antimicrobial substance synthesis.Additionally,ultra-high performance liquid chromatography coupled with high resolution mass spectrometry was applied to characterize the antimicrobial compounds synthesized by Z-14.Corn stover silage fermentation was performed using Z-14,and the microbial population,pH,and acid detergent fiber(ADF)content of the silage were subsequently analyzed.The results showed that Z-14 exhibited significant antagonistic activity against various types of pathogenic fungi and bacteria,with a wide inhibition spectrum.The Z-14 genome contains 8 clusters of antagonistic substance synthesis genes and 2 unknown clusters of antibiotic synthesis genes.The fermentation supernatant of Z-14 contained antimicrobial substances including fengycin,iturin A,surfactin,bacillaene,difficidin,bacillibactin,rhizocticin and macrolactin H.Z-14 significantly reduced aerobic decay of silage corn stover after bagging,decreased the growth of fungi,effectively slowed down the rise of pH,reduced the content of acidic detergent fiber,and improved the fermentation quality of corn stover.This study has laid a solid material and theoretical foundation for effectively preventing aerobic decay of corn silage straw and improving the quality of silage feed.关键词
